A Study to Learn How Different Amounts of the Study Medicine Called PF-08103402 Are Tolerated and Act in the Body in Healthy Adults or Adults With Mild To-moderate Asthma

The purpose of this study is to learn about the safety of a new study medicine called PF-08103402 in healthy adults (do not have disease) and or in adults with mild-to-moderate asthma. This is the first time the study medicine is being given to people. For Parts A, B, C, D and F, the study is seeking participants who: * Are healthy (do not have disease) males or females who can no longer have children, * Are 18 to 65 years old, * Have a body mass index (BMI) of 16 to 32 kilograms per meter squared and a body weight of more than 50 kilograms (110 pounds). Body mass index is a way to measure body fat by using a person's height and weight For Part A (optional group or cohort 3: Japanese participants only): * A body weight of more than 45 kilograms (100 pounds). * Have 4 biological Japanese grandparents who were born in Japan. For Part E only: * Adults with a documented history of asthma (confirmed by a doctor) for at least 12 months before entering the study. * Have a body mass index (BMI) of 16 to 35 kilograms per meter squared and a total body weight of more than 50 kilograms (110 pounds). The study has six parts: Part A, Part B, Part C, Part D, Part E and Part F. The study medicine will be taken as a suspension or tablet by mouth 1 time a day (except in Parts B and E where it will be taken 1 time a day for 14 days) at the study clinic. The study will help understand: * how the body processes the study medicine in healthy participants (Parts A and B), * how much of the study medicine gets into the bloodstream and if food affects the amount of study medicine in the blood in healthy participants (Part C), * how the study medicine is broken down and leaves the body in healthy participants (Optional Part D), * how the study medicine is processed in adults with mild-to-moderate asthma (Optional Part E), * if taking the study medicine together with another medicine affects how each medicine is processed by the body in healthy participants (Optional Part F). Participants will take part in the study for about 10 weeks (Parts A and F), 12 weeks (Part B), 9 weeks (Parts C and D), and 16 weeks (Part E). During this time, they will have 2 study visits at the study clinic and up to 28 overnight stays (Part A), 18 overnight stays (Parts B and E), 10 overnight stays (Part C), 11 overnight stays (Part D), and 16 overnight stays (Part F). The study team will also call participants 1 time over the phone at the end of the study to assess how they are doing. Study measurements will be taken by body examination, monitoring side effects, blood and urine tests, heart tests (ECG), vital signs (blood pressure, pulse and respiratory rate), questionnaires (Parts C and E), stool samples (Part D only), and breathing tests (Part E only).

Turkish Version of the BARSE

Background Physical inactivity is known to rank fourth among modifiable mortality risk factors globally and plays a central role in the etiology of noncommunicable diseases. In the context of physical activity, self-efficacy is a multidimensional construct, primarily divided into task self-efficacy and barrier self-efficacy. While task self-efficacy is crucial for initiating physical activity, barrier self-efficacy-the individual's belief in their capability to maintain exercise in the face of various physical, social, or psychological obstacles-is essential for long-term adherence. The Barrier-Specific Self-Efficacy Scale (BARSE), developed by McAuley et al., is specifically designed to measure this barrier self-efficacy dimension. A comprehensive literature review revealed no published Turkish validity and reliability study of the BARSE. Signed permission has been obtained from the original author to conduct this cultural adaptation. Objectives The primary aim of this study is to investigate the validity and reliability of the Turkish version of the BARSE among healthy young adults, thereby contributing a standardized, culturally adapted instrument to the scientific community. Study Design and Procedures This methodological study will conduct at the Faculty of Health Sciences, Hatay Mustafa Kemal University, between September-October 2026 with an enrollment of 133 healthy adult participants. The BARSE is a unidimensional scale consisting of 13 items. Participants are asked to rate their confidence in continuing to exercise under various challenging situations on a scale from 0% (not at all confident) to 100% (highly confident) in 10% increments. To establish construct validity and examine related health behaviors, participants will also complete several standardized tools: the Self-Rated Abilities for Health Practices Scale (SRAHP), the Exercise Benefits/Barriers Scale (EBBS), the Self-Perceived Barriers for Physical Activity Questionnaire (SPBPA), the Short Form Health Survey (SF-36), and the International Physical Activity Questionnaire-Short Form (IPAQ-SF). Statistical Analysis The language adaptation will conduct using the standard translation and back-translation method. Content validity will be evaluated using the Davis technique. Construct validity will be assessed through factor analysis and correlation coefficients. For reliability analyses, the scales will be administered twice with a 15-day interval. Reliability will be determined by calculating Cronbach's alpha coefficient, corrected item-total correlations, and the Intraclass Correlation Coefficient (ICC). Potential differences between test and retest measurements will be evaluated using appropriate statistical tests.

Feasibility, Acceptability, and Safety of a Drug-Free Nasal Spray in Frontline Healthcare Workers

This study looked at whether frontline healthcare workers could use a drug-free nasal spray twice a day for 8 weeks, whether it was safe, and whether they found it acceptable. The spray used in the study (Profi Nasal Spray, Akita Biosciences) was a cosmetic nasal hygiene product made from ingredients already used in nasal and topical products. Healthcare workers at a public hospital in New York City were randomly assigned to use the spray twice a day for 8 weeks or to receive no spray. The study asked: * Feasibility: Could busy healthcare workers use the spray as directed over 8 weeks? * Safety: Were side effects more common in people who used the spray than in people who did not? * Acceptability: How did users feel about the spray, and did they stay in the study? Every 3 days, participants answered a short online questionnaire about spray use, side effects, respiratory symptoms, and contact with patients who had COVID-19, influenza, or RSV. Spray use was checked by weighing the bottles at weeks 4 and 8. To better understand acceptability, about 20 spray users also took part in one-on-one interviews about what it was like to use the spray, including ease of use, fitting it into a daily routine, and anything that made it harder to use.

Cross-validating the iTBS-induced Hemodynamic Response From fNIRS and fMRI: a Concurrent iTBS-fMRI-fNIRS Study

Intermittent theta-burst stimulation (iTBS) targeting the left dorsolateral prefrontal cortex (DLPFC) is well established in treating patients with major depressive disorder (MDD). However, the iTBS-induced hemodynamic response during or immediately after a full session of stimulation has not been fully investigated. Although several concurrent iTBS/functional Near Infrared Spectroscopy (fNIRS) and iTBS/functional magnetic resonance imaging (fMRI) studies have investigated the hemodynamic response during or immediately after left DLPFC iTBS, they revealed inconsistent findings in the stimulated DLPFC. Given that the variability in iTBS-induced hemodynamic response in the stimulated DLPFC could be modulated by various inter- and intra-individual factors, cross-validating the findings from both techniques (fNIRS and fMRI) in the same group of participants under the same conditions through a simultaneous fMRI/fNIRS setting enables a straightforward comparison and interpretation of results from each technique. However, to our knowledge, cross-validation of iTBS-related fNIRS and simultaneous fMRI data has not been conducted yet, even the implementation of a simultaneous iTBS/fMRI/fNIRS setup is technically feasible with the mutually compatible equipment. Therefore, the current study has two primary objectives: Firstly, to establish a novel and practical methodological pipeline for simultaneously conducting iTBS/fMRI/fNIRS. Secondly, to cross-validate the hemodynamic responses during and immediately after iTBS, respectively monitored by fNIRS and fMRI.

Trancranial Pulse Stimulation on Anterior Insula Cortex and Dorsal Anterior Cingulate Cortex

Transcranial pulse stimulation (TPS) is a newly developed non-invasive brain stimulation (NIBS) technique from Austria \& Germany with highly promising applicability in neuropsychiatric disorders. Clinical trials have shown a beneficial effect of TPS in patients with Alzheimer\'s disease and depression. Although TPS has the capability to noninvasively target deeper brain regions such as the dorsal anterior cingulate cortex (dACC) and anterior insula cortex (AIC), no TPS study has been conducted to investigate the feasibility and effectiveness of dACC or AIC stimulation. Here, a randomized, single-blind, sham-controlled clinical pilot trial is proposed to probe the effects of TPS over bilateral dACC and AIC on modulating cognitive, behavioral and emotional functions and functional connectivity of brain circuits.
