NCT07882654
A PHASE 2A, EXPLORATORY, RANDOMIZED, SINGLE-BLIND, PLACEBO-CONTROLLED STUDY TO EVALUATE THE EFFICACY OF WEIGHT LOSS INDUCED BY NKS-3 IN OBESE ADULTS.
This study is designed as a single-blind, placebo-controlled, randomized, parallel-group trial. Subjects will be randomized in a 2:1 ratio to receive either NKS-3 or matching placebo, with a total planned enrollment of 31 subjects. During the 90-day treatment period, subjects will self-administer the assigned blinded study treatment daily at home. Given the exploratory nature of the study, which is intended to assess the feasibility of and generate preliminary evidence supporting the study hypothesis, a single-blind design has been selected. This approach reduces the logistical and organizational complexity associated with implementing a double-blind design while maintaining blinding of study participants to minimize expectation bias. Considering the exploratory objectives of the study, the selected design is considered appropriate and is not expected to compromise the scientific validity or the safety of participants.

Effects of Tirzepatide on Gut Microbiota and Gastrointestinal Function After Drug Withdrawal in Obese Patients
This is an observational cohort study. The primary objective is to analyze the dynamic changes in gut microbiota and metabolic profiles in obese patients treated with Tirzepatide. A total of 80 participants will be enrolled: 40 normal-weight controls (NW group) and 40 obese patients receiving Tirzepatide as part of their routine clinical care (OB-T group). Fecal samples for 16S rRNA sequencing and blood samples for metabolic markers will be collected at baseline and after 3 months. Changes in gut microbiota diversity and composition will be correlated with improvements in glucose and lipid metabolism. As per the approved protocol, exploratory follow-up assessments of gastrointestinal symptoms and gastric emptying parameters will also be conducted post-treatment.

Effects of Pistachios on Postprandial Metabolic Responses in Adults With Overweight or Obesity
This randomized controlled crossover study will examine the acute effects of pistachio consumption on postprandial metabolic responses in adults with overweight or obesity. Participants will complete two experimental conditions in randomized order. In one condition, participants will consume 30 g of pistachios with a standardized meal, and in the other condition they will receive an isocaloric refined-carbohydrate control condition with the standardized meal. The study will evaluate postprandial insulin and glucose responses over approximately 3 hours after meal consumption. Additional outcomes will include continuous glucose monitoring measures, blood lipid responses, appetite and satiety ratings, and exploratory measures related to insulin sensitivity and gastrointestinal hormone responses. The study is designed to determine whether pistachio consumption alters acute metabolic responses compared with an isocaloric refined-carbohydrate control in adults with overweight or obesity.

Very-Low-Carbohydrate Diet for Prediabetes and Obesity
Prediabetes is a condition where blood sugar is higher than normal but not yet high enough to be called type 2 diabetes. It is common in adults who are overweight or obese, and without treatment it often progresses to diabetes over time. This study looks at whether following a very-low-carbohydrate diet for 12 weeks can help adults with prediabetes and obesity lose weight, reduce body fat, and improve how well their body responds to insulin, compared with standard dietary advice. Adults aged 30 to 65 with obesity and newly diagnosed prediabetes are invited to take part. Participants are randomly assigned to one of two groups: one follows a very-low-carbohydrate diet with support from a dietitian, and the other receives standard dietary advice without a specific carbohydrate limit. Both groups are followed for 12 weeks, during which their weight, body composition, blood sugar, insulin levels, cholesterol, and kidney function are checked. The study aims to find out whether the very-low-carbohydrate diet leads to greater improvements in weight, body fat, and insulin sensitivity than standard advice, and whether it is safe and practical to follow in everyday life. The findings may help doctors and dietitians offer more effective ways to prevent prediabetes from progressing to type 2 diabetes.

Obesity Awareness, Weight Status, and Lifestyle Behaviors Among Adolescents
This school-based cross-sectional study evaluated the relationship between obesity awareness, weight status, and lifestyle behaviors among adolescents aged 15-17 years. A total of 751 students from public and private high schools in Istanbul participated. Sociodemographic characteristics, dietary habits, physical activity, screen exposure, sleep duration, and obesity awareness were assessed using structured questionnaires, and standardized anthropometric measurements were performed. The study also examined whether obesity awareness differed according to body mass index category and whether family and socioeconomic factors were associated with obesity awareness and excess weight.
